‘These drugs have real potential to be reused … safe for suppressing appetite and reducing weight’
Study leader Chen Liu, PhD, assistant professor of internal medicine and neuroscience, said: “We have shown that there is a real potential for reuse of these drugs, which are already recognized as safe. Peter O’Donnell Jr. Brain An investigator of the Institute for Depression and Weight Loss.
Treatment of obesity usually focuses on improving eating habits and physical activity – but migraine medicine offers a scientific alternative.
Serotonin, a chemical messenger found in the brain and body, plays an important role in increasing appetite. But there are 15 different types of serotonin receptors and that’s why researchers have struggled to understand each serotonin receptor’s role in appetite.
This means that previous drugs that target certain individual receptors, including Fan-Phan and Lorcaserin (Belvik), have been withdrawn from the market because of side effects.
However, triptans, which work by targeting a different receptor — the serotonin 1B receptor (Htr1b) — haven’t been well studied in the context of previous appetite and weight loss, Dr. Liu says.
Triptans can cause significant weight loss in less than a month.
For the study, researchers tested six prescriptions of triptans in obese mice that were fed a high-fat diet for seven weeks.
The rats ate two of these triptans in roughly equal amounts, while the other four rats ate less. After 24 days, the rats were given a daily dose of furotriptan, which reduced their body weight by an average of 3.6%. The rats that were not given triptans gained an average of 5.1% of their body weight.
Dr. Liu and colleagues saw similar results when they applied devices to animals that gave them regular doses of furutriptan for 24 days.
“We found that these drugs, and in particular, can reduce body weight and improve glucose metabolism in less than a month, which is quite impressive,” says Dr. Liu.
Triptans are usually prescribed for short-term use during migraines so that patients may not have experienced long-term effects on appetite and weight in the past.
The triptans cause weight loss by targeting the serotonin 1B receptor.
To determine how furotryptin affects food intake and weight gain, the researchers engineered mice to lack either Htr1b or Htr2c. In mice without Htr1b, Frovatriptan could no longer reduce appetite or cause weight loss, whereas cutting Htr2c had no effect. This confirmed that the drug works by targeting the serotonin 1B receptor.
“This discovery could be important for drug development,” says Dr. Liu. attracted attention. ,
The team clearly identified which neurons in the brain are most important for Htr1b’s role in mediating appetite, penetrating a small group of cells inside the brain’s hypothalamus.
